City smashed six goals past Championship leaders Burnley on Saturday to book their trip to Wembley, where they will face the team placed second in the second tier.
The Blades will also be without Jason McAtee and Tommy Doyle, who are both on loan from the Etihad Stadium with the latter notching a fine late winner past Blackburn on Sunday.
Brighton scored five against Grimsby to continue their fine season, reaching the semi-finals for the first time since a 1-0 defeat to Man City in 2019.
The semi-finals will be played across the weekend of April 21- 22.
