Two tries from Cooper Coats, plus scores from Alex Russell, Jake Thiel and Matthew Owuru condemned Richie Pugh's side to a loss that is a massive blow to their hopes of reaching the quarter-finals.
Wales play Zambia this evening before facing Fiji in the final group game on Saturday. The top two teams qualify for the knockouts, with the Welsh side realistically now having to beat Olympic champions Fiji to make it. The Fijians beat Zambia 52-0 in their opening match.
The Welsh team found themselves 17-0 down at the break and simply had no answer to the Canadians who extended their lead in the second period of a torrid match for the Welsh in Coventry.
It comes a week after it was announced the Wales team is being pulled from the World Series going forward and replaced by Team GB, which will include the best players from England and Scotland.
Wales weren't the only side to suffer on the opening day of the competition as England men were stunned by a 34-0 defeat by Samoa.
Steve Onosai scored a first-half hat-trick with Vaafauese Apelu Maliko, Vaovasa Afa Sua and Faafol Falaniko adding to the score.
England will struggle to reach the last eight as they still have to play defending champions New Zealand in the group stage.
Scotland, meanwhile, eased to a 41-0 win over Tonga after South Africa easily defeated Malaysia 46-0.
