Chasing WA's first innings total of 300, Tasmania were reduced to 5-74 at Bellerive Oval with Jordan Silk unbeaten on eight and Tim Paine 11 not out.
Resuming at 1-5, the Tigers lost nightwatchman Nathan Ellis (three) and opener Tim Ward (17) within the opening half hour to be 3-25.
Western Australia then claimed Jake Doran (five) and the key wicket of Ben McDermott (27) before the end of the opening session.
Joel Paris (2-20) and Matthew Kelly (2-21) impressed with the ball for the competition leaders.
WA can guarantee their spot in the Shield final with victory in Hobart, and potentially lock up hosting rights depending on other results.
Tasmania are one of four states in second on the standings.
