First-choice right-back Reece James is battling a hamstring injury, while his replacement Jarell Quansah limped off with an ankle twist during the 2-0 victory against Panama.
Quansah's injury adds to existing concerns, as fellow full-back Tino Livramento had already withdrawn from the squad due to a calf issue.
Despite goals from Jude Bellingham and Harry Kane securing the win over Panama, Tuchel admitted worries about the fitness of James and Quansah for the upcoming matches.
England is set to face DR Congo in the round-of-32 tie in Atlanta on Wednesday, with Tuchel confident in finding solutions for the defensive challenges.
