Naomi Osaka's Australian Open title defence is over after losing to American Amanda Anisimova. (AAP)

American Amanda Anisimova has saved two match points to send defending champion Naomi Osaka crashing out of the Australian Open at Melbourne Park.

The 20-year-old set up a fourth-round meeting on Sunday with Australian world No.1 Ash Barty with a fearless performance to shock Osaka 4-6 6-3 7-6 (10-5) on Friday night.

Up 5-4 in the third set, Osaka had match points on Anisimova's serve but couldn't close it out with the world No.60 taking full advantage in the match tiebreaker.
