A man armed with a knife has been arrested in a regional town after another man's body was discovered on the side of a rural highway.

Police were called to Firebrace Street in Horsham, about 300km northwest of Melbourne, at 8.30pm on Monday after reports of a knife-welding man behaving erratically.

Officers negotiated with the armed man before he was arrested about 11pm and taken to hospital.

Homicide Squad detectives are expected to interview the 49-year-old Minyip man at a later stage after the discovery of another man's body hours earlier.

Deputy Commissioner Bob Hill told reporters on Monday that a yet-to-be identified man was discovered with serious injuries on the side of a the road.

"I was briefed in relation to a deceased person that was located in the western district, a male on the side of the road who (had) serious injuries," he said.

"That matter is subject to investigation. We have our specialists deployed to that location."

The man is believed to be about 60 years old and his cause of death is yet to be determined.

Homicide Squad detectives have deemed the death as suspicious.
