Normal EFL Cup protocol sees the two relegated clubs join at the second round stage but owing to West Ham's Conference League win, both Leeds and Leicester join Southampton in finding themselves facing early August action.
The 12 Premier League clubs not competing in Europe will join in round two, with the other eight Premier League clubs starting in round three.
Clubs will be split into two sections, with Leeds along with Yorkshire rivals Hull City, Sheffield Wednesday, Huddersfield Town, Barnsley, Harrogate Town, Doncaster Rovers and Bradford City all in the northern section of the draw.
Opening round fixtures will take place in the midweek following the opening weekend of EFL games. Leeds' league campaign will be revealed at 9am next Thursday, with the Premier League's fixtures coming out this Thursday, June 15, at 9am.
