The whole area from 48th to 56th Streets is shut down in both directions and residents have been told to avoid the area.
Videos show a huge blaze from the car wreckage near the intersection of South 52nd Street and O Street, outside the Barnes & Noble's Bookstore, while people are seen stumbling and fleeing the area.
Initial reports suggest the deadly accident happened around 10.45 pm local time after a car reportedly went through a red light, hitting another vehicle and then the two cars span out of control, crashing into a crowd of people on the pavement.
One witness told the Journal Star that a speeding car crashed into another vehicle attempting a U-turn at the junction.
One of the vehicles then rolled up the curb and crashed into the crowd.
The man, who declined to provide his name, said some of those injured were children.
The incident took place near the Americruise event, hosted by the Midwest Association of Car Enthusiasts (MACE), however, it was not clear if the event was linked to the crash.
It occurs most years and sees motorists flock to Lincoln over Memorial Day Weekend to show off their cars by cruising the city streets.
A description of the event said: "Every Memorial Day Weekend, hundreds of cars, bikes, and trucks flock to Lincoln, NE to cruise down O Street to show off their rides and do some hooning!"
The event organisers also warned spectators that there may be risks involved.
They said: "We want to remind everyone the police won't be a stranger to this event so please cruise at your own risk and be cautious to those around you.
"There will be large crowds all along O street, and we don't need anyone in a wreck or worse in the hospital."
Police are yet to release more information.
