Martinez's withdrawal reduced United to 10 men when they were 2-1 up at the time as they had made their five substitutions on a night Raphael Varane was also forced off through injury at half time.
Varane, Martinez, Luke Shaw, Scott McTominay and Marcus Rashford are all now sidelined through injury for United, who travel to Nottingham Forest on Sunday before their return leg in Seville next Thursday, which is three days before their FA Cup semi-final against Brighton.
Ten Hag on substitutions
Erik ten Hag claims he had no choice but to substitute key personnel last night.
After Varane's enforced withdrawal through injury at half time, Ten Hag removed Bruno Fernandes, Jadon Sancho and Anthony Martial for Christian Eriksen, Anthony Elanga and Wout Weghorst on the hour with United 2-0 up.
Elanga has not scored a club goal since February 22 last year and Weghorst emerged having tallied two goals in 20 United games - both of them rebounds.
Antony, another goalscoring threat who rattled the crossbar in the second half, also made way for Facundo Pellistri in the 81st minute.
